The 32-year-old holed out from the fairway for a spectacular eagle at the par 4 third hole to widen the gap between himself and nearest challenger Collin Morikawa.
Matsuyama made his first birdie of the day two holes later at the par 5 fifth.
It is a hole he could probably play blindfolded as he has remarkably made birdie or better the last 20 times he has played the 520-yard hole which is ranked the easiest on the course.
Matsuyama dropped a shot at the 532-yard par 4 seventh as he missed the green left and failed to get up-and-down to save par, but he earned that shot back at the very next hole with a birdie at the scenic par 3 eighth where players tee off over a long canyon.
He then closed out his front nine with another birdie at the par-5 ninth.
After nine holes, Matsuyama is -31 for the tournament with Morikawa four shots back.
Australian Cam Davis has continued his surge up the leaderboard to move into the top five.
The 29-year-old shot back-to-back nine-under par 64s in the second and third rounds, and is five-under par for the day through 11 holes to be -23 overall.
Adam Scott is three shots back from his compatriot also with seven holes remaining, while Jason is -13 for the tournament with two holes left to play.
