Robert Toia produced a brilliant offload to put Mark Nawaqanitawase over for the first try as the Roosters took the lead.
James Tedesco produced the last pass to put over for a double and move him to 22 tries for the season and extended his lead for the most in the NRL.
Jye Gray produced a brilliant break to put Ashton Ward over for a long-range stunner to get the Rabbitohs back in the game.
A late tackle form Daniel Tupou on Isaiah Tass sparked an all-in melee with both sides rushing in.
Jamie Humprheys was sin-binned and placed on report for a hip drop tackle on Siu Wong as Souths were reduced to 12 men for 10 minutes.
James Tedesco came up with some silky hands to put Daniel Tupou over as the Roosters extended their lead.
Tedesco then put Connor Watson over for a long-range try in his 100th game for the club.
Nawaqanitawase soared high to catch a Sam Walker kick and seal a stunning hat-trick to all but seal a Roosters finals berth.
Tedesco then earned a penalty to allow Sam Walker to make it 30-6 heading into the final quarter.
